I want to cultivate Escherichia coli in a microtitre plate. Which conditions should I choose?

You should use 96-deepwell plates equipped with air-permeable sealing tape as a sterile barrier. The filling volume should not be greater than 500µL. Shake the plates with an orbital speed of 300 rpm and a shaking diameter of 50 mm. With smaller shaking diameters you should increase the shaking speed. How can evaporation loss in a microtitre plate be prevented?

On the one hand by using a shaking incubator with moisture control adjusted to 80-90 % humidity. On the other hand by choosing the appropriate sterile barrier. Additional data is available from the following publication:

Is there a difference between round and square wells?

Generally, the oxygen transfer rate is better in a 96-square well microplate with the corners functioning as baffles [q.v. Hermann R., Lehmann M., Buechs, J., Biotech. Bioeng. (2003); Vol. 81, No. 2, pp 178-186].
Also available are new shaped wells (Flowerplate), which can further elevate the oxygen transfer. This kind of plate is only available in a 48-well format at the moment:
